On October 18, the Institute of Life Sciences and the Sixth Affiliated Hospital of Sun Yat-sen University published the latest research results on rectal cancer in Cell Report Medicine.Based on DNBelab C4 and Stereo-seq, a single-cell library platform developed by Huada, the team revealed the dynamic changes of tumor microenvironment and drug resistance in rectal cancer under neoadjuvant chemotherapy.
Colorectal cancer is one of the high incidence malignant tumors in China, and its treatment plan has been a hot research area.At present, adjuvant chemotherapy with fluorouracil and oxaliplatin, or neoadjuvant chemotherapy, can significantly reduce tumor recurrence rate, protect organ function, reduce surgical trauma, and avoid side effects of radiotherapy.However, the mechanism of neoadjuvant chemotherapy on tumor microenvironment is unclear, and the relationship between tumor microenvironment and neoadjuvant chemotherapy efficacy is still to be studied.
In the study, the team recruited 29 patients with rectal cancer, sampled them before and after neoadjuvant chemotherapy, and sequenced them in single-cell and spatial groups.The team divided the samples into three groups: CR (complete remission), PR (partial remission) and NR (no significant remission).
The study reveals the complex regulatory relationship between tumor microenvironment and neoadjuvant chemotherapy.On the one hand, the team found for the first time that neoadjuvant chemotherapy had a significant effect on multiple components in the microenvironment and was associated with efficacy.Among them, fibroblasts (CAF) were remarkably remodeled and differentiated into new cell subtypes, enriching and activating effector T cells and dendritic cells, thus inhibiting tumor progression.On the other hand, the team found that another class of CAF cells in the microenvironment promotes tumor growth and migration, resulting in worse therapeutic outcomes.
Qin Pengfei, co-author of the paper and associate researcher of Huada Institute of Life Sciences, said: "We have been able to systematically and comprehensively study the tumor microenvironment under new adjuvant chemotherapy.With the addition of spatial group information, we have increased the dimension of field of view and improved the methodology in studying the spatial distribution, interaction, signal regulation and cell differentiation.”
Recent studies have shown that neoadjuvant chemotherapy is similar to neoadjuvant chemotherapy in colorectal cancer, avoiding the complications of radiotherapy and reducing the pain of patients.Our samples and studies provide important support and evidence for studying the relationship between tumor microenvironment and therapeutic effect and improving treatment regimen.Lin Hongcheng, co-author of the paper and deputy director of the Sixth Affiliated Hospital of Sun Yat-sen University, said.
